Campbell\nAssistant Professor\nUniversity of Alberta \n \nAt the heart of the Campbell group's research program lies the\nextra ordinarily useful family of fluorescent proteins (FPs) which\nincludes the green FP (GFP) from Aequorea jellyfish and\nits numerous homologues of va rious color from coral. All FPs share\nthe ability to autonomously generat e a visible wavelength\nfluorophore from a sequence of 3 amino acids locat ed in the center\nof their barrel-like structures. FPs are credited with s parking the\nexplosive and continuing growth in the popularity of live cel l\nfluorescence microscopy\; culminating in the 2008 Nobel Prize in\nChemi stry being awarded to 3 pioneers in this area\; Shimomura\,\nChalfie\, and Tsien.\nSince its inception in 2003\, the Campbell research group has\nwo rked to mould (that is\, engineer) FPs into improved tools for\naddressing fundamental questions in life science. For example\, we\nhave developed s everal improved alternatives to some of the\ntraditionally preferred hues of FPs [1\,2\,3] and exploited their\nimproved properties in new applicati ons. Some of our novel\napplications include the use of FPs for investigat ing peptide\nstructure in vivo [4]\, dual-FRET imaging of caspase activati on\nduring apoptosis [5]\, and a FP-based assay of nucleoside\ntransporter activity (with Joe Casey) [6]. In this seminar I will\ndiscuss some of ou r most recent efforts to create FP variants that\nhave physical or spectra l properties that are either improved\nrelative to existing variants or ar e completely novel and have not\nbeen previously reported. As the title of this seminar indicates\,\nthis discussion will focus on 3 aspects of FPs\ ; chain topology\,\nsurface properties\, and chromophore chemistry.
