DESCRIPTION:Engineering precise synthetic circuits and what it can teach us

 Engineering synthetic biological systems holds the potential for valuable 
 applications\, ranging from environmental remediation to biomedicine. It c
 an also deepen our fundamental understanding of natural phenomena\, by bui
 lding gene circuits and biological systems from the bottom up. Synthetic g
 ene circuits are able to perform a variety of useful tasks\, but despite b
 eing made from well-characterized biological components\, their precision 
 and robustness are usually much lower than their natural counterparts. Our
  hypothesis for this difference is that synthetic circuits have so far bee
 n designed mostly without accounting for the inherent stochastic gene expr
 ession in living cells.\n\nTo demonstrate this\, I will show how harnessin
 g principles from stochastic theory to redesign a classic synthetic oscill
 ator (the repressilator\, which helped jump-start the field of synthetic b
 iology) helped produce oscillations that rival those of natural clocks. I 
 will then describe how this motivated us to engineer hundreds of oscillato
 rs using a newly developed method for screening dynamical phenotypes\, thu
 s allowing us to explore areas that are beyond our current theoretical und
 erstanding. Finally\, I will showcase applications of synthetic circuits t
 o study natural systems\, such as measuring bacterial growth in the gut of
  live mice and measuring protein degradation in single cells\, and discuss
  the biological insights learned by engineering precise synthetic circuits
 .\n
